Monoamine oxidases: old friends hold many surprises

چکیده

The two forms of monoamine oxidase (MAO), A and B, continue to be of major interest to biochemists, pharmacologists, neurologists, and gerontologists. Despite intensive study for more than half a century, unexpected and unique properties of these enzymes continue to come to light. Recent studies have centered on their kinetic mechanism, their unique predilection for substrates related to the neurotoxic tertiary amine MPTP, and their putative role in aging and in the etiology of neurodegenerative diseases. New and potent inhibitors of MAO A and MAO B continue to be developed because of their potential use in clinical medicine. Some are effective in the picomolar range but MAO B from different mammalian species shows remarkable differences in sensitivity to these agents.Singer, T. P., Ramsay, R. R. Identification of Intracellular Sources Responsible for Endogenous Reactive Oxygen Species Formation

The endogenous reactive oxygen species ("ROS") formation is associated with many pathologic states such as inflammatory diseases, neurodegenerative diseases, brain and heart ischemic injuries, cancer, and aging. The purpose of this study was to investigate the endogenous sources for "ROS" formation in intact isolated rat hepatocytes, in particular, peroxisomal oxidases, monoamine oxidase, xanth...

Recent developments in the regulation of monoamine oxidase form and function: is the current model restricting our understanding of the breadth of contribution of monoamine oxidase to brain [dys]function?

Historically, much of the focus on monoamine oxidases and their substrates has been in the area of depression and the monoamine neurotransmitters serotonin (5-hydroxytryptamine), noradrenaline, and to a lesser extent, dopamine. With both forms of monoamine oxidase (A and B), the production of hydrogen peroxide as a byproduct of the reaction between the monoamine oxidases and their monoamine sub...

Monoamine Oxidases—New Vistas. Advances in Biochemical Psychopharmacology. Volume 5

This volume consists of a collection of papers on the classical mitochondrial monoamine oxidase and other amine oxidases presented at the Festschrift for Dr. H. Blaschko at Cagliari, Sardinia, 1971. In it the results of many investigations on the biochemistry, pharmacology and possible function of these enzymes over the past several years have been brought together. Much of the book is devoted ...

Activity-Based Probes for Studying the Activity of Flavin-Dependent Oxidases and for the Protein Target Profiling of Monoamine Oxidase Inhibitors**

High profile: new activity-based protein profiling (ABPP) probes have been designed that target exclusively monoamine oxidases A and B within living cells (see picture; FAD=flavin adenine dinucleotide, FMN=flavin monodinucleotide). With these probes it could be shown that the MAO inhibitor deprenyl, which is in clinical use against Parkinson's disease, shows unique protein specificity despite i...
